Boss challenge mode contains 5 stages where the players will be able to challenge bosses directly. The list below provides the disadvantages players will receive during each battle.

| Xia Hou Dun | Items cannot be used during the battle |
| Cao Ren | Cao Ren has maximum armor mode during the battle |
| Zhang Liang Zhang Bao | All supers cost 3 times as much |
| Zuo Ci | Players will be poisoned during the battle |
| Gan Ning | Archers will appear randomly on both sides during the battle and fire arrows onto the battle area |
| Lu Xun | Players' speed will be reduced during the battle |
| Xu Zhu | No supers may be used during the battle |
| Lu Meng | Lu Meng's speed and aggression are both increased |
| Xia Hou Yuan | 5 bolts of lightning will be targeted on players every 5 seconds |
| Si Ma Yi | Players will be dizzied on every wakeup |
| Meng Huo | Meng Huo kills players instantly upon any successful attack |
| Yan Liang & Wen Chou | Both bosses regenerate health constantly during the battle |
| Dong Zhuo | Players are permanently confused during the battle (4 and 6 switched, 8 and 2 switched) |
| Cao Cao & Sun Quan | Dual boss battle |
| Lu Bu | Only combos above 11 hits begin dealing damage to Lu Bu |
